How can I show ads inside a topic and before the leave a reply. Also how can I customize how to show ads inside a topic. Say show ads after three paragraph or before first paragraph, etc.
I have gone through the available ads location but can't see any

You should use the Post List option for the "Ad Banner Locations". See the screenshot below.
BTW, you can create different Ads for topics, posts, etc.
Don't forget to configure the "Topic List - Banner displaying frequency" option in the Dashboard > Forum > Settings > Addons > Advertisements admin page.
